Surface Plasmon Resonance (SPR)
This method involves attaching one interacting partner to the surface of a sensor chip, and then passing a sample containing the second interaction partner over the surface.
Binding of molecules to the sensor surface generates a response which is proportional to the bound mass, and can be monitored in real time.
Applications
-
Interaction specificity
-
Interaction kinetics and affinity
-
Drug discovery
-
Antibody characterization
Requirements
-
Biacore tubes with penetrable caps (various sizes)
-
A Biacore sensor chip with the required surface modification
-
Means to attach on of the interacting partners to the chip surface
Advantages
-
Real time measurements
-
High sensitivity
-
Quantitative method
-
Relatively high throughput
-
Sensor chips can be reused
