Sample concentration

In the far-UV region (<260 nm) - proteins have an extinction coefficient of about 7,000 M-1 cm-1. Therefore, the protein concentration should be approximately 0.2 to 0.4 mg/ml.

In the near-UV region (>260 nm) - the absorbance of proteins depends mainly on the concentrations of the aromatic amino acids, particularly tryptophan. Therefore, the protein concentration should be approximately 0.45 mg/ml for proteins with a high tryptophan content, and ≥2.5 mg/ml for proteins with low tryptophan content.
Sample volume
The volume of the sample is determined by the size of the cuvette.
